Suppose you claim “regular gifts out of income” of 4,000 and also give a gift of 2,000. Can you roll over the unused Annual Exemption of 1,000 to the next year? To put it another way, is roll-over affected by also claiming regular gifts out of income?

It's an annual exemption. Unused amounts can be rolled over, but only to the next year.
With a pinch of salt take what I say, but don't exceed your RDA
